Gene targeting strategy for B-hIL11 mice.  

The exons 1~5 of mouse Il11 gene that  encode the the full-length protein were replaced by human IL11 exons 1~5 in B-hIL11 mice.

mRNA expression analysis

Strain specific analysis of IL11 gene expression in WT and hIL11 mice by RT-PCR.

Mouse Il11 mRNA was detectable in splenocytes of wild-type (+/+) and heterozygous B-hIL11 mice (H/+). Human IL11 mRNA was detectable only in heterozygous B-hIL11 mice but not in wild-type mice.

Protein expression analysis

Strain specific IL11 expression analysis in heterozygous B-hIL11 mice by ELISA.

Serum was collected from wild-type mice (+/+) and heterozygous B-hIL11 mice (H/+), and analyzed by ELISA with species-specific IL11 ELISA kit. Mouse IL11 was detectable in wild-type mice and heterozygous B-hIL11 mice. Human IL11 was exclusively detectable in heterozygous B-hIL11 mice but not in wild-type mice. ND: not detected.
