mRNA expression analysis
RT-PCR analysis of IL12A and IL12B gene.
The human IL12A but not mouse IL12A mRNA was detectable in splenocyte of the homozygous B-hIL12A/hIL12B mice.
The human IL12B but not mouse IL12B mRNA was detectable in thymus cell of the homozygous B-hIL12A/hIL12B mice.
Protein expression analysis
Strain specific IL12(heterodimer p70) expression analysis in homozygous B-hIL12A/hIL12B mice by ELISA.
Serum was collected from WT (+/+) and homozygous B-hIL12A/IL12B mice (H/H) mice stimulated with LPS in vivo (n=3), and analyzed by ELISA with species-specific IL12 ELISA kit. Mouse IL12 (p70) was detectable in WT mice. Human IL12 (p70) was exclusively detectable in homozygous B-hIL12A/IL12B mice (H/H) mice but not in WT mice. ND: Not detectable.
